簡介:福建農林大學碩士學位論文多花水仙若干品種類型親緣關系的分子生物學研究姓名吳菁華申請學位級別碩士專業(yè)生化與分子生物學指導教師呂柳新20040401福建農林火學壩_學位論義摘要水仙II一2是II一1的同源三倍體提供了佐證。4水仙單染色體微分離與微克隆1水仙I1的根尖為材料,制備出用于微分離的染色體標本,利用顯微操作系統(tǒng),采用粘取法成功分離出I1中一個特殊的中著絲粒的染色體。2利用LAPCR方法成功地對該單染色體進行了體外擴增,獲得了片段大小為5002500BP的水仙中著絲粒染色體DNA庫。3用PGEMTEASYVECTOR克隆PCR產物,構建了單染色體的DNA質粒文庫。隨機分析部分重組子,發(fā)現(xiàn)文庫的插入片段大小為6002500之間。關鍵詞水仙;過氧化物酶同工酶;隨機擴增多態(tài)性DNA;熒光原位雜交;染色體微分離單染色體DNA質粒文庫;

簡介:國內闡書分類號國際圖書分類號東北龍膽農學博士學位論文密級GENTIANAMANSHURICAKITAGAWA生物學研究王臣導師胡寶忠教授申請學位級別;農學博士專業(yè)名稱作物遺傳育種論文提交日期2005年4月25門答辯日期2005年5月所在單位東北農業(yè)大學授予學位單位東北農業(yè)大學學位授予日期答辯委員會主席/CA5馳栽培。。。,,,查些些墜蘭垡堡皇墼絲耋。。。。。,。一苦苷含量有明顯差異,泰康我省西部栽培的高于巴彥我省中部栽培的龍膽苦營含量在不同個體和不同變異型之間含量差異顯著,進行高含量育種將是切實可行的。綜合分析本文的研究結果,作出以下結論1東北龍膽在抗旱性、抗病性、光合效率、經(jīng)濟系數(shù)以及藥材質量上明顯優(yōu)于粗糙龍膽,其單株生物量較低的不足可以通過提高栽培密度彌補,因此,東北龍膽應該作為按GAP進行生產的酋選種質。2栽培東北龍膽的遺傳組成為雜合體,與粗糙龍膽白然雜交是產生雜合體的原因之一通過雜交手段可以將粗糙龍膽和東北龍膽的優(yōu)良性狀組合在一起,創(chuàng)造新品種。另外,東北龍膽栽培群體的豐富的變異同時表現(xiàn)在形態(tài)上和有效成分含量上,可以通過對高含量個體的篩選實現(xiàn)栽培種的優(yōu)質化。3在沙壤土上栽培東北龍膽,各項指標均優(yōu)于在草甸土上栽培,說明黑龍江省中西部沙壤土地帶適于發(fā)展東北龍膽的栽培生產。這一結論給我們的啟示是原生地綜合條件對于有效成分的積累不一定是最好的,在研究提高產量的栽培技術時更應重視高質量栽培條件的研究。4將龍膽苦菅含量、折干率、可溶性糖和單株重四項指標結臺在一起,栽培龍膽的最佳采收期應在8月末至9月20日之間。關鍵詞東北龍膽;栽培;生物學;龍膽苦苷;發(fā)育形態(tài)學II

簡介:山東大學碩士學位論文粘細菌SOCE9882子實體毒理學研究及生物學功能的初步評價姓名唐慧申請學位級別碩士專業(yè)微生物學指導教師李越中20050513山東大學碩士學位論文摘要纖維堆囊菌是粘細菌中唯一能降解纖維素的屬,粘細菌是革藍氏陰性、單細胞、滑動細菌,它主要存在于土壤、糞便、腐爛的植物材料以及活的或死的樹干上。作為~種土壤中廣泛存在的細菌,它可以在營養(yǎng)貧瘠的條件下形成子實體,大量細胞共周協(xié)作生活,這一特點在原核生物中獨一無二。而且,粘細菌子實體在形態(tài)和結構方面有不同程度的復雜性,這一特點與藥用真菌的子實體有相似之處。到目前為止,關于纖維堆囊菌的研究工作主要集中在它能產生豐富的次級代謝產物這~方面,而對其子實體的應用開發(fā)國內外未見文獻報道。本文的土要目的就是探索粘細菌中纖維堆囊菌SORANGIUMCELLULOSUM子實體作為藥用菌材料的可行性和藥用前景。下面是論文工作的簡要介紹首先。對纖維堆囊菌SOCE9882子實體提取物進行毒理學分析,包括經(jīng)口急性毒性試驗、AME8試驗、小鼠骨髓嗜多染紅細胞微核試驗、小鼠精子畸形試驗。通過實驗可以得出結論在本次實驗條件下,受試物為實際無毒物質,未顯示有遺傳毒性作用,可以初步認為纖維堆囊菌SOCE9882子實體提取物是安全可靠的。其次,本文對纖維堆囊菌SOCE9882子實體提取物的藥理學活性進行了初步探索。主要做了一個方面的嘗試,即按照活性多糖的研究方法對粘細菌子實體熱水粗提物進一步的分離,同時對乙醇回流萃取的脂類物質按照極性的不同進行了初步分離。對分離到的部分組分進行細胞毒型抗癌藥物和癌化學預防藥物的篩選,結果表明組分6、7具有選擇性殺傷腫瘤細胞的活性,組分3、6具有肯定的抗突變效應麗組分7抗突變作用顯著。最后,運用紅外技術對篩選出來的組分進行測定,建立紅外光譜指紋圖譜,為探索纖維堆囊菌予實體提取物的生物活性作用機理提供依據(jù)。綜上所述,本論文工作為粘細菌子實體進一步的藥物研發(fā)打下了較為堅實的基礎。關鍵詞粘細菌;纖維堆囊菌;予實體;毒理學分析;生物學活性;紅外光譜分析。4

簡介:廈門大學博士學位論文條紋斑竹鯊養(yǎng)殖生物學、核型和線粒體DNA序列的研究姓名謝仰杰申請學位級別博士專業(yè)海洋生物學指導教師蘇永全20080701條紋斑竹鯊養(yǎng)殖生物學、核型和線粒體DNA序列的研究7條紋斑竹鯊的二倍體染色體數(shù)為2N104,在已知染色體核型的軟骨魚類中為二倍體數(shù)最多的種類之一,核型公式為2N40M26SM8ST30T,染色體總臂數(shù)NF170,其中第3L對染色體帶有隨體。8對采自廈門和湛江海域的條紋斑竹鯊的線粒體DNA控制區(qū)和CYT6基因進行PCR擴增和測序,分析了線粒體DNA控制區(qū)、CYT6基因和TRNA‰基因全序列GENBANK序列號EU363740~EU363752。結果表明,TRN礦。基因長度為69BP,控制區(qū)長度為1094~1096BP,CYT6基因全序列長度為1146BP。TRNA№基因非常保守,未發(fā)現(xiàn)堿基變異。所測30個個體中,控制區(qū)只有4個多態(tài)位點,5種不同的單倍型;CYT6基因全序列共有10個多態(tài)性位點,7種單倍型。CYT6基因的1146個堿基編碼381個氨基酸,氨基酸鏈共有3個變異位點,4種單倍型,廈門群體具有4種單倍型,湛江群體只有1種單倍型。綜合CYT6基因和控制區(qū)序列,2個條紋斑竹鯊群體共有LO種單倍型,其中廈門群體有9種單倍型,湛江群體僅4種單倍型。研究結果表明條紋斑竹鯊在控制區(qū)和CYT6基因序列上的遺傳多樣性均較低,廈門群體的遺傳多樣性水平高于湛江群體。’關鍵詞條紋斑竹鯊;養(yǎng)殖生物學;胚胎發(fā)育;核型;線粒體DNA

簡介:1094055分類爭Q96授予學位單位代碼J10434研究生學號20040222山柬裳薯大學碩士學位論文大猿葉蟲不同地理種群的生態(tài)適應性和山東種群生物學特性的研究研究生董宇奎學科專業(yè)農業(yè)昆蟲與害蟲防治研究方向昆蟲生態(tài)與害蟲綜合治理學院植物保護指導教師李照會教授2007年6月18日Ⅲ陸MⅫY啷隅剮GO.≯霉一胤脅糍一霉MC噸始M訓耐.1寶勰一N唧山東農業(yè)大學碩士學位論文中文摘要大猿葉蟲COLAPHELLUSBOWRINGIBALY屬鞘翅目COLEPTERA,葉甲科CHRYSOMELIDAE,無緣時甲屬COLAPHELLUS,是我國十字花科蔬菜上的一種重要食葉害蟲。本文以其為試驗材料,闡述了大猿葉蟲山東種群的生物學特性,系統(tǒng)地對不同地理種群大猿葉蟲的生態(tài)適應性進行了研究,包括不同的光周期和溫度對大猿葉蟲不同地理種群滯育誘導的作用、不同地理種群大猿葉蟲抗寒能力和低溫生存能力,以及光照和溫度對滯育后種群抗寒性的影響。主要結果如下1光周期和溫度的滯育誘導對于江西種群,當光周期的暗期長度為12~24H時,有效地防止滯育的發(fā)生;而當暗期長度短于12H則誘導不同程度的滯育,4~8H的暗期長度明顯誘導了較高的滯育率。黑龍江種群和山東種群不存在光周期反應。溫度本身能夠誘導滯育的發(fā)生。當溫度低于20℃時,所有的個體均迸入滯育;隨著溫度的逐漸上升,三種群的滯育比例逐步降低。28℃時,江西和山東種群的滯育率分別為22%和92%,而黑龍江種群的所有個體仍處于滯育狀態(tài)當溫度達30℃,江西和山東種群滯育率分別降至5%和78%時,黑龍江種群滯育率降為94%,出現(xiàn)非滯育個體。2大猿葉蟲耐寒性研究隨著緯度的升高,大猿葉蟲耐寒性逐漸增強。黑龍江、山東和江西三種群過冷卻點均于一月份達到最低值,分別為23.7“C、22.3℃和20.7℃,種群間差異顯著。試驗結果表明滯育與非滯育成蟲的低溫生存能力存在一定的差異一20℃時,三種群滯育成蟲的半致死時間分別為0.99H、0.75H和O.64H,而非滯育成蟲的半致死時間相對較短,分別為0.50H、0.34H和0.29H,滯育成蟲的耐寒能力明顯強于非滯育成蟲,北方種群耐受低溫的能力明顯強于南方種群。其它低溫下的情況也顯示了同樣的結果。各種群的過冷卻點都低于當?shù)馗髟缕骄鶞囟龋蕉上x可以保持較高的越冬存活率。4“C低溫馴化LOD后,大猿葉蟲黑龍江種群成蟲的耐寒能力無變化,
