簡介:近年來減毒細(xì)菌活載體因其能夠刺激機(jī)體產(chǎn)生針對(duì)活載體及其攜帶的外源抗原的體液、細(xì)胞免疫和局部粘膜免疫受到人們的廣泛關(guān)注特別是近期研究結(jié)果證明許多活細(xì)菌載體能夠作為DNA疫苗的運(yùn)送載體可以通過口服減毒細(xì)胞進(jìn)入機(jī)體從而誘導(dǎo)產(chǎn)生針對(duì)特定抗原的各種免疫應(yīng)答但迄今為止對(duì)活疫苗載體與宿主免疫系統(tǒng)間相互作用規(guī)律及其調(diào)控機(jī)制知之甚少這方面的免疫生物學(xué)研究對(duì)于研制新型基因工程疫苗將會(huì)提供重要的理論依據(jù)且對(duì)相關(guān)疾病的免疫防制有著重要的指導(dǎo)價(jià)值因此我們構(gòu)建了表達(dá)綠色熒光蛋白GFP的減毒鼠傷寒沙門氏菌闡明其在體內(nèi)、外感染試驗(yàn)的動(dòng)態(tài)分布及它與抗原呈遞細(xì)胞的相互作用規(guī)律這為沙門氏菌疫苗載體及相關(guān)基因工程疫苗的研究奠定了理論基礎(chǔ)1表達(dá)綠色熒光蛋白的重組減毒鼠傷寒沙門氏菌的構(gòu)建與鑒定2重組減毒鼠傷寒沙門氏菌感染動(dòng)力學(xué)3重組減毒鼠傷寒沙門氏菌與抗原呈遞細(xì)胞的相互作用綜上所述該研究構(gòu)建了表達(dá)綠色熒光蛋白的鼠傷寒沙門氏菌通過靜脈注射和口服途徑記錄了此細(xì)胞在體內(nèi)一些臟器的分布規(guī)律及誘導(dǎo)的抗體的變化結(jié)果顯示細(xì)菌數(shù)量及抗體效價(jià)變化均有一定的規(guī)律而且細(xì)菌數(shù)量與抗體效價(jià)之間有一定的相關(guān)性在體外用細(xì)胞系和腹腔巨噬細(xì)胞驗(yàn)證了此細(xì)菌具有感染細(xì)胞的能力在體內(nèi)感染實(shí)驗(yàn)中檢測(cè)了靜脈注射途徑肝臟、脾臟中巨噬細(xì)胞和樹突細(xì)胞的感染率同時(shí)檢測(cè)了口服后3天腹腔巨噬細(xì)胞的感染狀況結(jié)果顯示在體內(nèi)巨噬細(xì)胞和樹突細(xì)胞很快地?cái)z取了該重組菌這為起始保護(hù)性免疫反應(yīng)奠定了基礎(chǔ)

簡介:山東農(nóng)業(yè)大學(xué)博士后士學(xué)位論文蘋果角質(zhì)層的生物學(xué)和虎皮病的發(fā)生姓名張?jiān)暾?qǐng)學(xué)位級(jí)別博士后士專業(yè)果樹學(xué)指導(dǎo)教師束懷瑞2001121內(nèi)部結(jié)構(gòu)已經(jīng)被破壞。在表皮細(xì)胞及下皮細(xì)胞周圍也有有大量電子密致物,電子密致物進(jìn)入細(xì)胞時(shí)伴有小囊泡的形成,細(xì)胞內(nèi)部結(jié)構(gòu)包括質(zhì)體被破壞。貯藏至20周時(shí),晚采果仍然沒有發(fā)生虎皮病,果皮細(xì)胞結(jié)構(gòu)完整,仍然可以見到質(zhì)體及線粒體。超微結(jié)構(gòu)中僅見表皮細(xì)胞上方有大量電子密致物,而表皮細(xì)胞下方只有少量電子密致物,且未或僅小部分接觸細(xì)胞。外層下皮細(xì)胞周圍未見有下移電子密致物,細(xì)胞內(nèi)部結(jié)構(gòu)完好,葉綠體結(jié)構(gòu)完整??傊?,角質(zhì)層中的電子密致物在蘋果虎皮病發(fā)生過程中可向內(nèi)層表皮細(xì)胞、下皮細(xì)胞移動(dòng),從外部破壞細(xì)胞,使細(xì)胞解體,產(chǎn)生褐變。推測(cè)角質(zhì)層中蠟層的消失及角質(zhì)層電子密致物的下移與小法尼烯的積累和氧化有關(guān)。研究表明在貯存過程中早采和晚采國光和紅星蘋果角質(zhì)層中的共軛三烯含量與虎皮病的發(fā)生存在不相關(guān)現(xiàn)象,進(jìn)一步證明了以共軛三烯含量來表示僅法尼烯氧化產(chǎn)物說明虎皮病的發(fā)生程度存在弊端。而RFC值與虎皮病的發(fā)生密切相關(guān)。RFC值可以表示角質(zhì)層中抗氧化水平。晚采和DPA處理的果實(shí)的抗氧化活性分別顯著高于早采果和未處理果,前者的RFC值也分別顯著高于后者。修正了A一法尼烯與共軛三烯的比值RFC值用于虎皮病預(yù)測(cè)的數(shù)學(xué)模型。模型可用于貯存過程中蘋集虎皮病的預(yù)測(cè)。通常在RFC值低于10時(shí)蘋果出庫至常溫時(shí)虎皮病即會(huì)發(fā)生弋關(guān)鍵詞蘋果;角質(zhì)層;蠟層;超微結(jié)構(gòu);虎皮??;電子密致物;旺法尼烯;共軛三烯II

簡介:中國科學(xué)技術(shù)大學(xué)碩士學(xué)位論文同步輻射小麥引起的生物學(xué)效應(yīng)血源與基因工程Α干擾素抑制貯脂細(xì)胞增殖及膠原合成的比較研究姓名趙凌申請(qǐng)學(xué)位級(jí)別碩士專業(yè)細(xì)胞生物學(xué)指導(dǎo)教師顧月華200141中國科學(xué)技術(shù)人學(xué)預(yù)上學(xué)位論文第一部分同步輻射對(duì)小麥引起的生物學(xué)效應(yīng)摘要研究了不同劑量的同步輻射軟X射線源輻照小麥種子后產(chǎn)生的生物學(xué)效應(yīng),以期從細(xì)胞和分子水平蛋白質(zhì)、酶及DNA等上揭示同步輻射軟X射線源對(duì)生物體的誘變效應(yīng)及其作用機(jī)制,為其應(yīng)用于輻射育種提供理論依據(jù)。I我們進(jìn)行了以下研究I檢測(cè)小麥幼苗細(xì)胞的脂質(zhì)過氧化作用及保護(hù)酶活性??梢钥闯?,隨著同步輻射軟X射線劑量的增加,脂質(zhì)過氧化產(chǎn)物丙二醛MDA的含量也隨之增加;在保護(hù)酶系統(tǒng)中,過氧化氫酶CAT、過氧化物酶POD、超氧化物歧化酶SOD的活性變化不表現(xiàn)出同步性,表明三者之間存在相互協(xié)調(diào)的關(guān)系,處理組葉綠素含量及葉綠體中關(guān)鍵的保護(hù)酶抗壞血酸過氧化物酶APX的活性始終低于空白對(duì)照組。結(jié)果表明同步輻射軟X射線輻照對(duì)于小麥細(xì)胞保護(hù)酶活性及脂質(zhì)過氧化作用的影響很大。II采用單細(xì)胞凝膠電泳方法檢測(cè)了同步輻射軟X射線源對(duì)DNA造成的單鏈損傷,通過測(cè)定DNA遷移效應(yīng),表明DNA損傷程度與輻照劑量呈顯著的正相關(guān)性。1ID采用TUNEL方法來進(jìn)行原位細(xì)胞凋亡檢測(cè),并結(jié)合DAPI熒光染色,表明同步輻射軟X射線源輻照小麥種子可以使之產(chǎn)生不同程度上的細(xì)胞凋亡。通過統(tǒng)計(jì)細(xì)胞凋亡率,也表明損傷程度與輻照劑量成『F相關(guān)性。以上實(shí)驗(yàn)表明,作為一種新的誘變?cè)?,同步輻射軟X射線輻照麥類作物種子的生物學(xué)效應(yīng)顯著,它不僅能夠?qū)π←溂?xì)胞遺傳物質(zhì)造成損傷同時(shí)也能對(duì)相關(guān)的體內(nèi)保護(hù)酶系統(tǒng)產(chǎn)生調(diào)節(jié)作用,因此應(yīng)用同步輻射進(jìn)行農(nóng)業(yè)育種將有、極大的發(fā)展?jié)摿颓熬啊?關(guān)鍵詞同步輻射軟X射線源;小麥細(xì)胞;脂質(zhì)過氧化;保護(hù)酶系統(tǒng);單細(xì)胞凝膠電泳;細(xì)胞凋亡
