簡介:丁基苯酞(3NBUTYLPHTHALIDE,NBP),是從芹屬植物的種子中提取的有效成分,2002年11月由國家食品藥品監(jiān)督管理局批準的治療缺血性腦卒中的國家一類新藥。但存在嚴重的不良反應(yīng),如使患者血糖升高,肝功能異常以及胃腸道反應(yīng)等。因此,鄭州大學化學系合成了更加安全有效的抗腦缺血的藥物NBP類似物布羅佐噴鈉【SODIUM±5BROMO2ΑHYDROXYPENTYLBENZOATE,5溴2(Α羥基戊基)】苯甲酸鈉,商品名BROZOPINE(BZP)。目前BZP已通過Ⅰ期臨床試驗。高血壓是腦卒中最重要獨立危險因素,而高鹽飲食是高血壓重要環(huán)境因素。因此,本研究通過建立鹽敏感高血壓大鼠模型,觀察BZP對鹽敏感高血壓大鼠神經(jīng)運動功能、血壓、腦、心臟和腎臟功能等的影響,進而探究BZP對鹽敏感高血壓大鼠腦卒中的防治作用其機制。第I部分鹽敏感高血壓大鼠腦卒中模型的建立目的通過高鹽飲食,建立鹽敏感高血壓大鼠(DAHLSSHYPERTENSIVERATS)腦卒中模型。方法12只7周齡雄性鹽敏感大鼠(DAHLSSRATS)和12只7周齡雄性SS13BN大鼠,適應(yīng)環(huán)境1周,隨機分為對照組和模型組,每組12只。其中對照組為SS13BN大鼠,正常飲食(03%SALT5L79DIET);模型組為DAHLSS大鼠,高鹽飲食(8SALTAIN76ADIET)。定期監(jiān)測血壓、體重、神經(jīng)功能評分(1次3D)。記錄大鼠的死亡率和腦卒中的發(fā)生率。48天后,檢測血漿中過氧化氫酶(CAT)、谷胱甘肽過氧化物酶(GSHPX)活力及一氧化氮(NO)的含量;取腦、心臟和腎臟HE染色,觀察病理學變化。結(jié)果1高鹽飲食對DAHLSS高血壓大鼠體重的影響高鹽飲食第12天,模型組大鼠體重明顯低于正常組(P2高鹽飲食對DAHLSS大鼠血壓的影響高鹽飲食12天,與正常組相比,模型組大鼠收縮壓(SBP)、平均壓(MAP)和舒張壓(DBP)明顯高于正常范圍(P3高鹽飲食對DAHLSS高血壓大鼠神經(jīng)運動功能的影響31高鹽飲食對DAHLSS高血壓大鼠神經(jīng)功能評分的影響模型組在高鹽飲食第12天,大鼠平衡木、肌力評定、BEDERSON以及綜合評定等神經(jīng)功能評分升高(P32高鹽飲食后DAHLSS高血壓大鼠腦卒中的發(fā)生率根據(jù)大鼠高鹽飲食后運動功能改變情況,我們將綜合評分≥3分定義為腦卒中。高鹽飲食第12天,917神經(jīng)功能綜合評分≥3分,說明高鹽飲食誘導(dǎo)鹽敏感高血壓大鼠腦卒中模型成功。并且隨著高鹽飲食攝入時間的延長,大鼠腦卒中的發(fā)生率逐漸增加,腦卒中癥狀也逐漸加重。4高鹽飲食對DAHLSS高血壓大鼠存活率的影響高鹽飲食48天期間模型組大鼠存活率(667)明顯下降。NBPK96MGKG組大鼠存活率明顯提高(917)。結(jié)合死亡前大鼠明顯的神經(jīng)行為學異常以及死亡大鼠腦組織視交叉附近可散在梗死灶及明顯水腫等,判定腦卒中是其死亡的主要原因。5神經(jīng)學評分與血壓的相關(guān)性分析模型組大鼠平衡木評分、肌力評定、BEDERSON評分及綜合評分與大鼠血壓均顯著相關(guān),神經(jīng)功能評分與收縮壓(SBP)相關(guān)系數(shù)分別為R1208373(P6高鹽飲食對DAHLSS大鼠臟器的損傷作用模型組腦組織細胞體積縮小,細胞核固縮,胞質(zhì)透亮,著色變淺,且不均勻,出現(xiàn)許多空泡,嚴重者出現(xiàn)核破碎,細胞排列疏松紊亂,間隙變大;腎小球萎縮、消失;纖維化、硬化和玻璃樣變性;間質(zhì)纖維組織增生和炎癥細胞浸潤;心肌細胞排列紊亂、細胞間隙明顯增大,肌纖維有斷裂,細胞核排列不整齊大小不均一。說明高鹽飲食可造成大鼠腦、心臟和腎臟等重要臟器的損傷。7高鹽飲食對DAHLSS高血壓大鼠氧化應(yīng)激水平的影響與對照組相比,模型組大鼠高鹽飲食后血漿中CAT活性和GSHPX活性顯著降低(P結(jié)論高鹽飲食可誘導(dǎo)鹽敏感高血壓大鼠腦卒中的形成。第II部分布羅佐噴鈉對鹽敏感高血壓大鼠腦卒中的防治作用及其機制目的探究布羅佐噴鈉(BROZOPINEBZP)對鹽敏感高血壓大鼠腦卒中的防治作用,從而為臨床應(yīng)用提供相關(guān)依據(jù)。方法72只7周齡雄性鹽敏感大鼠和12只7周齡雄性SS13BN大鼠,適應(yīng)環(huán)境1周,隨機分為正常組、模型組、BZP(075,3,12MGKG)組、BRNBP105MGKG組和NBPK96MGKG組,每組12只。對照組給予正常飲食(SS13BN大鼠,03SALT5L79DIET)和其余劑量組給予高鹽飲食(DAHLSSRATS,8SALTAIN76ADIET)。其中BZP12MGKG與BRNBP105MGKG、NBPK96MGKG為等摩爾劑量。高鹽飲食2周開始給藥,給藥方式為尾靜脈注射,每天一次,連續(xù)4周,正常組和模型組給予等體積的生理鹽水。在此期間監(jiān)測血壓、體重和神經(jīng)功能評分,每7天測一次。并記錄大鼠腦卒中的發(fā)生率和大鼠的存活率。給藥28天后,檢測血漿中過氧化氫酶(CAT)、谷胱甘肽過氧化物酶(GSHPX)活力及一氧化氮(NO)的含量;取腦、心臟和腎臟HE染色,觀察病理學變化。結(jié)果1BZP可增加DAHLSS高血壓大鼠體重高鹽飲食3周后,模型組大鼠與正常組相比體重明顯降低(P2BZP并不引起DAHLSS高血壓大鼠血壓的降低高鹽飲食2周后(給藥前一天),與正常組相比,模型組收縮壓(SBP)、平均壓(MAP)和舒張壓(DBP)明顯高于正常范圍(P3BZP對DAHLSS高血壓大鼠神經(jīng)運動功能的影響31運動功能評分高鹽飲食2周,大鼠均出現(xiàn)不同程度的運動協(xié)調(diào)功能障礙,平衡木評定、肌力評定、BEDERSON以及綜合評定等神經(jīng)功能評分升高(P32BZP可降低DAHLSS高血壓大鼠腦卒中的發(fā)生率根據(jù)大鼠的運動功能損傷情況,綜合評分≥3分則被認為該大鼠已形成腦卒中。高鹽飲食第7天,模型組有833的大鼠發(fā)生腦卒中。而BZP(075,3,12MGKG)組在給藥第7天腦卒中發(fā)生率與模型組相比明顯下降(583,583,167VS833)且呈現(xiàn)劑量依賴性。且BZP12MGKG降低大鼠腦卒中發(fā)生率效果優(yōu)于BRNBP105MGKG組和NBPK96MGKG組。4BZP可提高DAHLSS高血壓大鼠的存活率高鹽飲食期間,模型組大鼠存活率為583,BZP(075,3,12MGKG)組大鼠存活率分別為833,917,100。說明BZP能劑量依賴性地顯著提高大鼠存活率。結(jié)合死亡前大鼠明顯的神經(jīng)行為學異常以及死亡大鼠腦組織視交叉附近可散在梗死灶及明顯水腫等,判定腦卒中是其死亡的主要原因。與模型組相比,BZP(075,3,12MGKG)組大鼠存活時間明顯延長【(4125±205)天、(4125±260)、)天、(42±0)天VS(3942±454)天】。5BZP可提高CAT、GSHPX的活性,降低NO的含量與正常組相比,模型組大鼠高鹽飲食后血漿中CAT活性(P6組織形態(tài)學改變模型組大鼠腦細胞體積縮小,細胞核固縮,核質(zhì)比減小,胞質(zhì)透亮,著色變淺,不均勻,出現(xiàn)許多空泡,嚴重者核破碎,細胞排列疏松紊亂,間隙變大;腎小球萎縮、消失;纖維化、硬化和玻璃樣變性;間質(zhì)纖維組織增生和炎癥細胞浸潤;心肌細胞排列紊亂、細胞間隙明顯增大,肌纖維有斷裂,細胞核排列不整齊,不均一。BZP(075,3,12MGKG)組可顯著減輕腦、心、腎等重要臟器的;效果優(yōu)于NBPK96MGKG與BRNBP105MGKG組。表明BZP對鹽敏感高血壓大鼠所致腦,心臟和腎臟的損傷具有保護作用。結(jié)論BZP對鹽敏感高血壓大鼠腦卒中的有顯著防治作用。1BZP可顯著改善鹽敏感高血壓大鼠腦卒中引起的運動功能障礙;2BZP對鹽敏感高血壓大鼠所致腦、腎臟和心肌損傷具有明顯的保護作用;3BZP對鹽敏感高血壓大鼠腦卒中的防治作用機制可能與提高鹽敏感高血壓大鼠機體抗氧化能力有關(guān)。

簡介:近年來,高血脂及其相關(guān)的心血管疾病已經(jīng)成為人類甚至伴侶動物的主要健康問題。許多的降血脂藥物用于高血脂的治療,比如他汀類、貝特類、依澤替米貝類及煙酸類藥物。然而,絕大多數(shù)的藥物價格昂貴且有一定的不良效果。因此,開發(fā)一種低毒、便宜、安全、高效并可長期使用的藥物顯得十分必要。本研究主要探討阿司匹林丁香酚酯(AEE)對高血脂的預(yù)防和治療作用。1、AEE對高血脂的預(yù)防作用SD雄性大鼠被用來評估AEE對高血脂的預(yù)防效果。通過對大鼠飼喂6周的高脂飼料來建立高血脂的動物疾病模型。飼喂高脂飼料的同時,大鼠同時給予一定量的藥物。根據(jù)大鼠每周的體重,對大鼠進行灌胃給藥,AEE的給藥劑量分為高中低三個組,分別為低劑量組18MGKGDAY,中劑量組36MGKGDAY和高劑量組54MGKGDAY。在實驗中,10MGKGDAY的辛伐他汀和20MGKGDAY的羧甲基纖維素鈉被分別用作陽性和陰性對照藥物。實驗結(jié)束后,大鼠的血脂水平存在明顯差別。54MGKGDAY的AEE可顯著地降低甘油三脂、低密度脂蛋白和總膽固醇,表明AEE對高血脂有很好的預(yù)防作用。同時,大鼠體重的增長速度在AEE組明顯降低。2、AEE對高血脂的治療作用通過給予WISTA大鼠8周的高脂飼料建立高血脂動物疾病模型來評估AEE對高血脂的治療效果。在實驗中,AEE分為高中低三個劑量,分別為18、36和54MGKGDAY。為了比較AEE和其前體化合物之間的差異,與中劑量AEE摩爾量相等的阿司匹林、丁香酚及阿司匹林丁香酚(摩爾比11)也設(shè)計在實驗中。05%的羧甲基纖維素鈉(20MGKG)作為溶劑對照,其給藥體積與藥物組大致相等。辛伐他?。?0MGKGDAY)被用于陽性對照藥物。大鼠被分為10組,分別為空白、模型和八個藥物治療組。給藥5周后,血清的血脂水平通過自動生化儀檢測,檢測指標包括甘油三脂、膽固醇、低密度脂蛋白和高密度脂蛋白。在本實驗中,54MGKGDAY的AEE可顯著地增加高密度脂蛋白的水平P肝、胃和十二指腸的病理學檢查結(jié)果顯示,大鼠在給予高脂飼料和藥物后肝組織的病理學變化發(fā)生較大改變。胃和十二指腸無明顯變化。3、AEE代謝組學研究伴隨著血脂水平的升高,高血脂是心血管疾病的重要風險因子。本實驗中,通過飼喂大鼠高脂飼料的方法建立高血脂疾病模型。高靈敏的UPLCQTOFMS用于血漿分析。主成分分析法(PCA)和正交偏最小二乘判別法(OPLSDA)用于挖掘大鼠代謝的變化。通過SPLOT對潛在的生物標記物進行確認。大鼠血漿中與高血脂相關(guān)的22個生物標記物被篩選出來。除此之外,METPA被用于代謝通路的相關(guān)分析。實驗結(jié)果表明AEE抗高血脂的效果可能來自對脂質(zhì)代謝、能量代謝及氨基酸代謝的調(diào)節(jié)作用。結(jié)論在SD雄性大鼠中給予54MGKGDAY的AEE六周能夠有效地預(yù)防高血脂,并且其效果強于辛伐他汀。給藥五周之后,血脂指標在實驗中存在顯著的差異。在WISTAR大鼠中,治療高血脂的最佳劑量確定為54MGKGDAY,給藥時間為五周。AEE抗高血脂的效果可能來自對脂質(zhì)代謝、能量代謝及氨基酸代謝的調(diào)節(jié)作用。

簡介:背景高原病是發(fā)生于高原低氧環(huán)境中的一種特發(fā)性疾病。高原低壓性缺氧是致病的主要因素。目前,防治急性高原病的措施與方法有階梯適應(yīng)、藥物和吸氧等,但它們都存在不同的缺點和問題?,F(xiàn)階段,解決高原缺氧的技術(shù)手段主要有增氧和增壓兩大類。增壓的方法是通過增加大氣壓力,在氧氣體積百分比不變的情況下來提高氧分壓。增壓的裝備主要有便攜式增壓氣艙(袋)、高原輕便加壓艙、便攜式高原單兵增壓氧艙、加壓面罩、單兵高原增氧呼吸器等?,F(xiàn)有裝備存在結(jié)構(gòu)比較復(fù)雜、體量偏大、造價較高、需要專業(yè)人員操作、使用成本高或機動性差、不便攜帶、沒有流量壓力控制裝置、密閉性不好或增壓效果不十分理想等缺點。而由于高原地區(qū)氣候惡劣、嚴寒缺氧、地形復(fù)雜、道路狹窄崎嶇、保障能力相對差等特點,因此,研制體積小、重量輕、實用性強、安全方便的小型供氧裝備就顯得尤為迫切。方法基于增壓的原理,我們在專利“便攜式高原急救頭盔裝置”的基礎(chǔ)上,綜合運用物理、電子、機械和材料等相關(guān)知識,進行了一系列較大的改進設(shè)計,研制出了一種新的便攜式高原增壓頭盔,并在大型低壓艙模擬高原環(huán)境(海拔分別為3500M和5000M)檢測佩戴使用頭盔前后人體的經(jīng)皮血氧飽和度(PERCUTANEOUSOXYGENSATURATION,SPO2)和心率(HEARTRATE,HR)的變化(頭罩內(nèi)部壓強范圍分別為(0410)KPA、(0414)KPA、(0416)KPA);在高原現(xiàn)場(拉薩,海拔3650M)檢測其對急性高原病癥狀評分(按我國國家軍用標準進行評判)的改善效果,以及佩戴使用頭盔前后急性高原病患者的SPO2和HR的變化(頭罩內(nèi)部壓強范圍為(0414)KPA)。結(jié)果建立了“便攜式高原增壓頭盔”的制作工藝路線,包括機箱和頭罩的結(jié)構(gòu)設(shè)計、模具加工、材料篩選等;完成了“便攜式高原增壓頭盔”的試制和操作使用規(guī)范;探索發(fā)現(xiàn)了高原增壓頭盔的頭罩內(nèi)部最大壓強在10KPA、14KPA、16KPA時具有相似的效果,且在14KPA時相對較為適宜。在模擬高原環(huán)境,佩戴使用高原增壓頭盔可明顯提高人體的血氧飽和度、降低心率。在高原現(xiàn)場,佩戴使用高原增壓頭盔可顯著提高人體的血氧飽和度、降低心率,且使用高原增壓頭盔后第二天,急性高原病患者的癥狀評分顯著降低,與對照組比較有顯著性差異。結(jié)論我們研制了一種體積小、重量輕、實用性強、安全方便的供氧裝備高原增壓頭盔,在模擬高原環(huán)境和高原現(xiàn)場均可緩解人體缺氧、促進高原習服。為改善高原缺氧、防治急性高原病和促進高原習服等提供了一套有效的裝具,為高原衛(wèi)勤保障提供了物質(zhì)基礎(chǔ)和技術(shù)支撐,這對于高原上缺乏藥物和供氧設(shè)施差的邊遠小散地區(qū)人員,在突發(fā)急性高原病時的現(xiàn)地急救、癥狀緩解和醫(yī)療后送途中的持續(xù)增氧治療等,具有重要的應(yīng)用價值。
