3、bstract Bayannur Mutton Sheep is a high meat performance of Sheep varieties. It is created by Mongolian Sheep and Merino Sheep through grading breeding methods. In order to reveal its reproductive performance and make an
4、 effective utilization of this precious resource-Bayannur Mutton Sheep, this research used Bayannur Mutton sheep as the experiment material. First, the polymorphism of candidate genes (GnRHR、FSHβ、FSHR、LHβ、LHR ) were dete
5、cted through SSCP technology. Then, differentially expressed research of GnRHR、LHR and FSHR genes was done in ovary and uterus tissues of single Bayannur Mutton Sheep & biparous Bayannur Mutton Sheep in different est
6、rous period (1 h/12 h/24 h). Besides, Differential expression study of FSHR、 LHR and FSHβ genes was also done through Real-time Quantitative PCR(RT-PCR) in the left and right ovarian. (1) The polymorphism of these five g
7、enes was detected through SSCP technology. The results showed that polymorphism was only found in the exon2 of FSHβ in the three amplified fragments and AA, BB, CC, AB, AC, and CC genotypes were determined in this region
8、. The results also verified that SSCP was caused by cytosine (C) to thymine (T) transition, and the polymorphic positions were 87 and 147. Furthermore, the two substitutions did not influence the sequences of amino acids
9、. The frequencies of AA, AB, AC, and CC genotypes in Bamei Mutton Sheep were 0.4733, 0.2667, 0.1533 and 0.0867 respectively. For Bamei Mutton Sheep, there was a significant effect of the gene on the yeaning number and th
10、e Least Square mean value of yeaning number of genotype AB was larger than that of AA genotype (0.2257 of one goat, P<0.05), and was larger than that of AC and CC genotypes 0.6446 and 0.3135 (P<0.05) respectively.
11、In conclusion, the allele A of FSHβ may play a role in increasing yeaning number of Bamei Mutton Sheep. The other 4 genes (GnRHR、 FSHR、 LHβ、 LHR) did not show any polymorphism. (2) The RT-PCR reaction system of Bayannur
