3、mancdbloodstemcellsthealphafetoprotein(AFP)wastestedbyimmunohistochemmistrytoinvestigatethechimericalgrowthofhumanlivercellsinthemice.ThemicethenwereinfectedbyhepatitisBvirus(HBV)theserumAFPalbumin(ALB)werequantitatively

4、testedinthecomparisonwithnmalnudemice.Results:theAFPwascontinuouslyexpressedinthelivercytoplasmofmiceatthe7th14th21stdaysfallowingthehumancdbloodstemcellsinjection.ThereweredenseHBsAgpositivecellsobservedinmiceliverInfec

5、tedwithHBVbyimmunohistochemistrywhichweresignificantdifferentfromthoseinthecontrol(P0.05).Conclusions:humancdbloodstemcellstransplantedintothemouselivercouldsurvivethereindifferentiateintohumanhepaticcellsthusfmingthechi

6、mericawhichwereabletobeinfectedbyHBV.PartTwo:Theestablishmentofhumanmousechimericausinghumanmyeloidstemcell.Objectives:toestablishahumanmousechimericabyinvitrotransplantationofhumanmyeloidstemcells.Methods:Humanmyeloidst

7、emcellsintheamountof1109permouseweredirectlyinjectedintothefetushepatictissuesofmicebysurgery.ThehumanserumAFPALBinthetransplantedmiceweretestedimmunohistochemistricallyat2nd4th6th12thmonthsafterbirthtoinvestigatethechim

8、ericalgrowthofhumanlivercellsinthemice.InadditionthemicewerealsoinfectedbyhepatitisCvirus(HCV).Results:ThehumanserumAFPcontentintransplantedmiceatagesof24612monthsweresignificantlyhigherthanthatofnmalmice.Theimmunohistoc

9、hemistricallypositivehepaticcellsofhumanAFPALBwereobservedatageof2nd12thmonths.ThemiceinfectedbyHCVshowedapositiverateof66.67%.Conclusions:Humanmyeloidstemcellscouldbechimerisedintomouseliverdifferentiateintohumanhepatic
