1、目的:本實驗以Fischer大鼠甲狀腺細胞系(FRTL)為研究對象,在體外從細胞和分子水平研究高糖毒性及AGEs對甲狀腺細胞甲狀腺球蛋白(TG)、甲狀腺過氧化物酶(TPO)mRNA表達的影響,初步探索除免疫機制外,糖毒性致甲狀腺組織損傷的機制。 方法:在體外用D-glucose與BSA共同孵育AGEs-BSA。將體外培養(yǎng)成功的FRTL細胞分為+TSH組(完全培養(yǎng)基中加TSH)和-TSH組(完全培養(yǎng)基中未加TSH),每組分別給予不
2、同濃度的D-glucose、AGEs及AGEs抗體刺激,在糖刺激組設(shè)置滲透組以排除滲透壓因素對細胞的影響,于24h、48h、72h分別收集細胞,使用實時熒光定量PCR測定TG、TPO mRNA的表達情況。 結(jié)果: ①12周后得到棕褐色產(chǎn)物AGEs-BSA。AGEs-BSA熒光光譜掃描:其激發(fā)波長為360nm,發(fā)射波長為450nm,AGEs-BSA的熒光值高于BSA,其濃度為52.446g/ml。 ②實驗中FRTL
3、細胞無污染,圓形較亮,成堆生長,密度適中,貼壁良好。 ③實時熒光定量PCR檢測TG、TPOmRNA的表達水平:在+TSH組與-TSH組:D-glucose刺激組及AGEs刺激組中,每個刺激時間中組間比較均有統(tǒng)計學(xué)意義(P<0.001);且在D-glucose刺激組,對照組與滲透組比較差別無統(tǒng)計學(xué)意義(P>0.05);+TSH組不同濃度D-glucose刺激24h、48h、72h后;15mmol/L組低于對照組、滲透組(TG、TP
